MD Anderson hosted a news conference with Nobel Prize winner Jim Allison, Ph.D. Allison was awarded the 2018 Nobel Prize in Physiology or Medicine for launching an effective new way to attack cancer by treating the immune system rather than the tumor. Allison is the first MD Anderson scientist to receive the world’s most preeminent award for outstanding discoveries in the fields of life sciences and medicine. “I’m honored and humbled to receive this prestigious recognition,” Allison said. “A driving motivation for scientists is simply to push the frontiers of knowledge. I didn’t set out to study cancer, but to understand the biology of T cells, these incredible cells that travel our bodies and work to protect us.” Allison started his career at MD Anderson in 1977, arriving as one of the first employees of a new basic science research center located in Smithville, Texas. He was recruited back to MD Anderson in November 2012 to lead the Immunology Department and to establish an immunotherapy research platform for MD Anderson’s Moon Shots Program. The prize recognizes Allison’s basic science discoveries on the biology of T cells, the adaptive immune system’s soldiers, and his invention of immune checkpoint blockade to treat cancer. Allison’s crucial insight was to block a protein on T cells that acts as a brake on their activation, freeing the T cells to attack cancer. He developed an antibody to block the checkpoint protein CTLA-4 and demonstrated the success of the approach in experimental models. His work led to development of the first immune checkpoint inhibitor drug. Ipilimumab was approved for late-stage melanoma by the U.S. Food and Drug Administration in 2011. His drug, known commercially as Yervoy, became the first to extend the survival of patients with late-stage melanoma. Follow-up studies show 20 percent of those treated live for at least three years with many living for 10 years and beyond, unprecedented results. Subsequent research has extended this approach to new immune regulatory targets, most prominently PD-1 and PD-L1, with drugs approved to treat certain types and stages of melanoma, lung, kidney, bladder, gastric, liver, cervical, colorectal, and head and neck cancers and Hodgkin’s lymphoma. They actually turn off the genes that cause cancer (oncogenes), and turn on or activate tumor suppressor genes—genes that fight cancer. His treatment strategy, which he refers to as "Personalized Gene Targeted Cancer Therapy," includes mapping the patient's entire cancer genome. This involves analyzing some 24,000 genes in order to identify the abnormal genes. Once they've determined which genes are involved in the cancer, drugs and supplements are identified to target those specific genes. Antineoplastons work on approximately 100 cancer-causing genes, but traditional oncology agents (including chemotherapy) may also be used, typically in combination with antineoplastons. This expanded direction of "personalized gene-targeted treatment" has permitted people who would otherwise be denied access to the still-unapproved antineoplastons to benefit from his treatment. Vice President Joe Biden announced plans in September 2016 to make it easier for cancer patients to get information about clinical trials that could help them and speed up the work of researchers seeking new cures. “Every day thousands of Americans hear that dreaded word, that C-word," Biden said in a Sept. 16 speech sponsored by MD Anderson and Rice University’s Baker Institute for Public Policy. "What happens next is a difference between life and death. If there's treatment there, then you know there's hope, but for some people, hope is a clinical trial." Biden's speech at Rice University took place just a few days after the 54th anniversary of President John F. Kennedy's historic speech about going to the moon at Rice on Sept. 12, 1962. "President Kennedy's words are still ringing in my ear: 'We are unwilling to postpone for a minute, a day, a week, a month,' " Biden said. "Every great doctor in here can tell you — I guarantee you — he's been asked by a patient, 'Doc, can you give me just one more week so I can walk her down the aisle?' 'Doc, my first grandbaby's coming. I need three months, doc. Just give me three months.' "This is about today," Biden said. "This is not about tomorrow." Biden knows firsthand just how urgent the mission is for the millions of people touched by cancer. He recalled losing his son Beau last year after a devastating cancer diagnosis and thanked the MD Anderson doctors and nurses who offered hope and helped his son fight for his life. MD Anderson is “probably the greatest cancer research hospital in the world,” he said. MD Anderson President Ronald DePinho, M.D., said the national Cancer Moonshot event at Rice was a proud moment for Houston and MD Anderson, which launched its own Moon Shots Program in 2012, inspiring the national movement. Biden’s speech means that “he stands with us in the fight against cancer,” DePinho said. “It will put the might of the federal government behind these efforts.” MD Anderson’s cancer clinical trial program is the largest in the nation. More than 9,400 participants were enrolled in clinical trials exploring innovative treatments in Fiscal Year 2015. These trials are also an important part of MD Anderson’s Moon Shots Program. President Barack Obama announced the White House Cancer Moonshot to accelerate cancer research during his 2016 State of the Union address. K.C. was initially diagnosed with stage 3 lung cancer in 2015. After receiving chemotherapy and radiation, her healthcare team was not optimistic—her cancer had spread to her lymph nodes and progressed to stage 4. K.C. sought a second opinion in Houston and began treatment of nivolumab. She responded to the immunotherapy immediately, and her recent scans shows no evidence of disease. His victorious battles with the United States government were centered around Dr. Burzynski's gene-targeted cancer medicines he discovered in the 1970's called Antineoplastons, which have currently completed Phase II FDA-supervised clinical trials in 2009 and has been given permission by the FDA to begin the final phase of FDA testing--randomized controlled clinical trials. When Antineoplastons are approved, it will mark the first time in history a single scientist, not a pharmaceutical company, will hold the exclusive patent and distribution rights on a paradigm-shifting medical breakthrough. Antineoplastons are responsible for curing some of the most incurable forms of terminal cancer. Various cancer survivors are presented in the film who chose these medicines instead of surgery, chemotherapy or radiation - with full disclosure of medical records to support their diagnosis and recovery - as well as systematic (non-anecdotal) FDA-supervised clinical trial data comparing Antineoplastons to other available treatments—which is published within the peer-reviewed medical literature. One form of cancer - diffuse, intrinsic, childhood brainstem glioma has never before been cured in any scientifically controlled clinical trial in the history of medicine. Dr. Burzynski resides and practices medicine in Houston, Texas. He was able to initially produce and administer his discovery without FDA-approval from 1977-1995 because the state of Texas at this time did not require that Texas physicians be required to adhere to Federal law in this situation. This law has since been changed. As with anything that changes current-day paradigms, Burzynski's ability to successfully treat incurable cancer with such consistency has baffled the industry. Ironically, this fact had prompted numerous investigations by the Texas Medical Board, who relentlessly took Dr. Burzynski as high as the state supreme court in their failed attempt to halt his practices. Likewise, the Food and Drug Administration engaged in four Federal Grand Juries spanning over a decade attempting to indict Dr. Burzynski, all of which ended in no finding of fault on his behalf. Finally, Dr. Burzynski was indicted in their 5th Grand Jury in 1995, resulting in two federal trials and two sets of jurors finding him not guilty of any wrongdoing. If convicted, Dr. Burzynski would have faced a maximum of 290 years in a federal prison and $18.5 million in fines. However, what was revealed a few years after Dr. Burzynski won his freedom, helps to paint a more coherent picture regarding the true motivation of the United States government's relentless persecution of Stanislaw Burzynski, M.D., Ph.D. Note: When Antineoplastons are approved for public use, it will allow a single scientist to hold an exclusive right to manufacture and sell these medicines on the open market—potentially leaving the pharmaceutical industry absent in profiting from the most effective gene-targeted cancer treatment the world has ever seen.

Armed with National Cancer Institute funding, Cecilia Williams, an assistant professor in the University of Houston's Biology and Biochemistry Department, is leading single-cell level research aimed at better preventing and treating colon cancer and two types of breast cancer. Williams is studying estrogen hormones and the mechanics of regulatory molecules called nuclear receptors. Williams believes estrogen can help combat inflammation in colon cells and 'turn off' cancer-promoting genes. "Estrogen receptor beta is the one that appears protective against colon cancer," said Williams. "So, if you can activate that one (nuclear receptor), specifically in the colon, then we could prevent or delay colon cancer development." Williams is currently working to better understand exactly how this is able to happen and is testing a hypothesis.

Conducting cancer research in the microgravity environment of the International Space Station offers unique insight into the genetic and cellular processes that can’t be duplicated on Earth. Luis Zea, a research associate at BioServe Space Technologies, explains that cells grown in space arrange themselves into three-dimensional groupings, or aggregates, which more closely resemble what happens in the body. Understanding how viruses cause oral cancer and how our bodies interact with these viruses is very important to preventing infection throughout the body, not just in the mouth. Oral cancer research at the College of Dentistry covers a wide range of focus areas including innovative early detection methods; rapid, non-invasive, gene expression based diagnosis of oral cancer, changes in cells that occur with aging and effects of DNA damage; study of the development of human papillomavirus (HPV) associated cancers at the molecular level; and understanding how stem cells regulate telomere length and how disrupted telomere maintenance can lead to tumor formation and metastasis. Research performed at the UIC College of Dentistry Center for Molecular Biology of Oral Diseases, are focused on development of new treatments for head and neck cancer patients, and of new molecular diagnostic tools to better assess clinical outcomes in patients at risk for oral cancer. • Keiko Watanabe’s lab has two main focuses: 1). the effects of periodontitis on prediabetes and T2DM and, 2.) early detection of oral cancer. Periodontitis is a chronic inflammatory disease that leads to loss of tooth supporting bone. Oral SCC is an aggressive epithelial malignancy that is the sixth most common neoplasm in the world today. This team is studying the precise mechanism responsible for the association between periodontitis, T2DM, and obesity, using animal model systems. To advance early detection of oral cancer, the team is applying various methods to determine if PTK6 can be used as a prognostic marker to identity premalignant lesions which ultimately become malignant. • Charles Zhou’s Lab is focused on molecular profiling of oral cancer. The lab’s primary research interest is to utilize high-throughput genomics and bioinformatics technologies to gain a better understanding of this disease process and to develop novel diagnostic tools for head and neck/oral squamous cell carcinoma (SCC) patients.

Dr Nathan Fowler is Associate Professor at the Department of Lymphoma/Myeloma, University of Texas, MD Anderson Cancer Center, Houston, US. He is board certified in medical oncology and internal medicine and is currently Co-Director of the Clinical Investigation and Translational Research Program, Team Lead of the Low Grade Lymphoma Research Group, and Team Lead of the CNS Lymphoma Research Group, all at the MD Anderson Cancer Center. He received his medical degree from the University of Texas Medical Branch in 2001, and remained there for his clinical internship and residency. He completed his clinical fellowship at Georgetown University in Washington in 2007. Dr Fowler has produced more than 60 peer-reviewed papers and congress abstracts, as well as contributing to several textbooks on lymphoma and its treatment. He has served as Principal Investigator for a number of investigator-initiated Phase I and Phase II trials focused on non-Hodgkin lymphoma, including conducting the largest study into the use of immunomodulatory drugs in combination for untreated low-grade lymphoma and the first Phase I trial to demonstrate the activity of a Bruton's tyrosine kinase (BTK) inhibitor in humans. His primary clinical research interest is to further develop novel therapeutic regimens utilizing biologic and immunomodulatory agents to benefit patients with low-grade non-Hodgkin lymphoma.
