In addition to a typical certificate of analysis (COA), we also test our antibiotics through a modified version of the Kirby-Bauer method, also known as the disk diffusion method. The test compares the antibiotic's ability to prevent bacterial growth to a common set of standards created by the Clinical Laboratory Standards Institute (CLSI) in order to assure its functionality at a certain concentration. As a control, we use a strain sensitive to the antibiotic to determine its effectiveness and a resistant strain to assure it is the correct substance. Each lot is tested with this procedure. So generally we have to find out if there is a microorganism or any infection in the urine or not. So putting a urine sample for culture in an aerobic medium, if there is no growth, then it is possible that there is infection. But if the urine has infection and suspecting the growth and the microorganism is there. So growth or no growth depends on the microorganism in the urine or the infection that is present in the urine. So no growth in the urine culture specifies that the urine does not have any infection or the patient may be on antibiotics suppressing growth of the microorganism, then only we can subject the urine to some tests like sensitivity of the bacterial growth, to which antibiotic they are sensitive otherwise is to see if it is acidic or alkaline, does it have RBC’s or casts. So we can subject the urine samples for a routine microscopy and we don’t have to do a culture. Even routine microscopy can give also information on the urine sample.

However, they can be identified as lactose negative or lactose positive by what color they ferment lactose in MacConkey agar. In this episode, Dr. Pamela explains what this means and discusses treatment options for a gram negative infection. Hint: gram negative infections often resolve on their own. Therefore, it is not always necessary to treat with antibiotics. Remember, it is always advisable to consult your local veterinarian when making these decisions.

In order to grow, cells need very specific environmental conditions such as food, energy, proper temperature and humidity. When growing cells in the lab, we have to create these conditions using culture media. Solid culture media is a mixture of agar and nutrients poured into Petri dishes. We can transfer cells to the Petri dishes where they will grow with proper incubation. Sterile technique is required to create culture media so you don't have unwanted microorganisms growing in your culture. In this video, we'll demonstrate the steps for preparing Brain Heart Infusion Agar. It's a general-purpose media used for a variety of bacterial and fungal species including pathogens like Streptococcus and Pneumococcus.

A time-lapse of the Kirby-bauer test done with E. coli and S. aureus. The Kirby-Bauer test uses Mueller-Hinton agar to test a bacterium's susceptibility to antibiotics by using discs that have antibiotics in it. Antibiotics can work in one of two ways: they either slow down growth (bacteriostatic) or kill the bacteria (Bactericidal). A lot of antibiotics target the thick peptidoglycan layer of the cell wall in gram positive organisms, which makes gram negative bacteria more resistant to these antibiotics. If the bacteria is susceptible to the antibiotic, it will create a zone of inhibition which is the area around the antibiotic disc where the bacteria did not grow. A larger zone indicates greater susceptibility. Cultures were grown at approximately 37 degrees Celsius.

Webinar: Fast Phenotypic Antibiotic Susceptibility Testing of Positive Blood Cultures The role of the microbiology laboratory in the processing of positive patient blood cultures has become more complicated with the increasing demand for rapid information to assist in the management of sepsis. Many laboratories have turned to using molecular systems that report identification and resistant marker results that supply actionable information but no susceptibility data. These systems have proven over time to be useful with the management of Gram positive bloodstream infections but vary in the ability to properly guide therapy choices for Gram negative bacteremia. In our institution, phenotypic antimicrobial susceptibility results are often necessary for optimization of therapy. This is particularly true due to emerging resistance mechanisms in Gram negative bacilli, such as AmpC and porin alterations. The Accelerate Pheno™ system is a truly unique and divergent technology that has the potential to produce final susceptibility results directly from a positive blood culture within 7 hours. This is a reduction in time to results from conventional susceptibility methods of 2-3 days. The Accelerate Diagnostics system was FDA cleared in February of 2017. Clinical trial and scientific presentations have shown the identification and MIC susceptibility results to be reliable and can be used to improve patient care. How to Diagnose and Treat TB|How to Read a Tuberculosis Skin Test Disclaimer:this video only for educational purpose Tuberculosis, or TB, is a disease (usually of the lungs) caused by Mycobacterium tuberculosis (M. tuberculosis) and is easily transmitted through the air when an infected person speaks, sneezes, laughs, sings, or coughs. The airborne particles then get inhaled and enter the exposed person’s lungs, where they multiply. Although TB is rare and highly treatable in the US 1 Schedule a TB test if you have been exposed to someone with tuberculosis. If you have recently been exposed to someone with active TB and believe there is a chance you might have contracted the disease, it is important to consult your health care provider immediately. There are 2 methods for TB testing: Skin test: The Tuberculin Skin Test (TST) requires injecting a protein solution sometime between 8 and 10 weeks after contact with an infected person. The patient must return to the medical provider 2 or 3 days later to have the skin reaction interpreted. Blood test: Although it's not as commonplace as the skin test, the TB blood test only requires a single doctor visit and is less likely to result in misinterpretation by a medical professional. It is the necessary option for anyone who has received the BCG vaccination, as the vaccine can interfere with the accuracy of the tuberculin skin test. If your TB test is positive, you will need to undergo additional testing. Health professionals will need to determine whether you have a latent TB (which is not contagious) or active TB disease before proceeding with treatment. Tests may include a chest x-ray and a sputum test 2 Begin immediate treatment for latent TB. If you test positive for latent TB, you should consult with your doctor about the best course of action. Although you don't feel sick with a latent TB, and it isn't contagious, you will probably be prescribed a course of antibiotics to kill the inactive TB germs and prevent tuberculosis from turning into an active disease. The 2 most common treatments are: Taking isoniazid daily or twice a week. The duration of the treatment is 6 or 9 months. Or, for those who can’t tolerate isoniazid, taking rifampin daily for 4 months. 3 Begin immediate treatment for active TB. If you test positive for active TB, it is essential that you begin treatment as soon as possible. Symptoms of active TB include cough, sputum production, fever, weight loss, fatigue, night sweats, chills and a loss of appetite. Nowadays, active TB is highly treatable with a combination of antibiotic medications, however the duration of treatment can be quite long, usually between six to twelve months. The most common medications to treat TB include isoniazid, rifampin (Rifadin, Rimactane), ethambutol (Myambutol) and pyrazinamide. With active TB, you will usually need to take a combination of these drugs, especially if you have a particularly drug-resistant strain. Patients with resistance to both isoniazid and rifampin should be monitored for 2 years after treatment. If you follow your treatment plan exactly, you should start to feel better within a matter of weeks and you should no longer be contagious. There is a little more to this book than just that, it has a little bit of dosing instructions, and some information on why its "safe" to use vet meds - but I am afraid that this book will soon be horribly out of date, as the FDA and other Alpha Agencies are talking about requiring prescriptions to buy veterinary medications. What would have made Do it yourself medicine more useful as a reference would be a chart on animal body-weight to human body-weight - especially as it says that determining proper dosage of the medicine is hard. Is the same dose you give a 200 pound pig the same dose you would give a 200 pound human? I do not know, and before I jab a needle full of veterinary grade antibiotics in my hindquarters I would like to know. It may be a moot point, as I have heard rumblings of requiring prescriptions for vet medicine which would close this "loop hole"* *Update - They did

Disinfectants are antimicrobial agents that are applied to non-living objects to destroy microorganisms that are living on the objects. Disinfection does not necessarily kill all microorganisms, especially resistant bacterial spores; it is less effective than sterilization, which is an extreme physical and/or chemical process that kills all types of life. Disinfectants are different from other antimicrobial agents such as antibiotics, which destroy microorganisms within the body, and antiseptics, which destroy microorganisms on living tissue. Disinfectants are also different from biocides — the latter are intended to destroy all forms of life, not just microorganisms. Disinfectants work by destroying the cell wall of microbes or interfering with the metabolism.

If you eat or drink something that is contaminated with the bacteria, the bacteria enter your body. They travel into your intestines, and then into your blood. The bacteria travel through the blood to your Lymph nodes, Gallbladder, Liver, spleen, and other parts of the body. Some persons become carriers of S. typhi and continue to release the bacteria in their stools for years, spreading the Disease. Typhoid fever is common in developing countries. Fewer than 400 cases are reported in the U.S. each year. Most cases in the U.S are brought in from other countries where typhoid fever is common. Symptoms Early symptoms include fever, general ill-feeling, and abdominal pain. High fever (103°F, or 39.5°C) or higher and severe diarrhea occur as the disease gets worse. Some people with typhoid fever develop a rash called "rose spots," which are small red spots on the abdomen and chest. Other symptoms that occur include: Abdominal tenderness Agitation Bloody stools Chills Difficulty paying attention (attention deficit) Delirium Fluctuating mood Slow, sluggish, lethargic feeling Weakness Exams and Tests A complete blood count (CBC) will show a high number of white blood cells. A blood culture during the first week of the fever can show S. typhi bacteria. ELISA urine test to look for the bacteria that cause Typhoid fever Fluorescent antibody study to look for substances that are specific to Typhoid bacteria Platelet count (platelet count may be low). Treatment Fluids and electrolytes may be given by IV (into a vein). Or you may be asked to drink uncontaminated water with electrolyte packets. Antibiotics are given to kill the bacteria. This lecture describes blood, urine and skin/soft tissue cultures, focusing on the types of media, sample collection processes, culture procedures, as well as speciation and susceptibility testing. Basic staining procedures, such as the simple stain, gram stain, spore stain, negative stain, and acid fast stain are briefly discussed. Here is the 2nd part of the "at home penicillium" culture test. I took varying amounts of liquid culture broth and put them on cut filter paper on nutriet agar plates that were covered with B. subtilis. I plated 4 experimental dishes vs one control dish with two 10 unit discs of penicillin. B. subtilis is a gram positive bacteria, and so should respond to pencillin. Unfortunately, I wasn't able to check the plates until 3 days after the initial plating, so I saw no rings of inhibition. It also adds new effects and attempts to eliminate the need for unnecessary drugs and effects resulting in a cleaner aid section. Non Survival This version adds new drugs and meds with positive effects like restoring HP, Giving you positive buffs and etc. Survival Hp is your overall health. If it's low, It doesn't mean that you've been physically hurt, it just shows you if you're healthy or not. Crippled: If you get crippled it means that you've been shot or stabbed or you've fell off a cliff. When you get crippled you need perform a surgery on yourself (or visit a doctor for a safer procedure) which will be painful and has some side effects. Surgery side effects include pain, blood loss, open wound and etc. Diseases act as normal but antibiotics are not the cure to all of them, They just cure infection. (Keep reading for more details) Medications (Limb) 1. Surgery kit: A kit that allows you to perform a emergency surgery on yourself. This procedure is painful and can cause: Pain (Your character gets annoyed and you feel the effect every few minutes) Blood Loss (You'll have less stamina and speed) Open wound (It can get infected and you'll have less damage resistance) 2. Pain Killer: A remedy for your pain. If over used it can be addictive. 3. Blood Pack: You can make up for your lost blood by using a blood pack. Warning it can be dangerous to use and there is a small chance that it'll cause heart attack. over using it will cause cardiac arrest. (You can use it 3 time within 24 hours) Cardiac arrest (You can do pretty much nothing about it, Just make sure your HP doesn't fall and you can recover over time.) Harvest Blood Kit: A kit that enables you to harvest your own blood. (adds mild blood loss for just a few minutes) Safe Blood Pack: It's your own blood and doesn't have any side effects like Cardiac arrest. 4.Bandages: There are 2 variants, Clean and Dirty. They both cure open wound but the dirty one can infect your wound. Horizon Patch: 2 more bandages, Sterile (also has HP bonus) and Used bandage (Which is just gross.) Fresh wound (Bandages will add fresh wound which is safe and will be healed after a while) 5. First aid kit: This is a kit that allows you to perform a much safer procedure and has less side effects. You have the skill to examine the drugs and use the right dosage to prevent side effects (Much harder to come by) Horizon Patch: Just helps you carry more drugs and when you use it you just get them back. Advanced Surgery Kit: Does whatever First aid Kit does without the patch. Medications (Disease) 1. Antibiotics: Cures infection. 2. Sleep aid: Cures insomnia. It is addictive so, moderation. 3. Anodyne: Cures fatigue. 4. Anti-parasitic: Cures parasite. 5. Stimulant: Cures Lethargy. 6. Energy Pills: Cures Weakness. Medications (HP) Normal HP restoring meds like stimpak or Curie's health thing and other things like water and food. Stimpak no longer cures damaged limbs. For a Hardcore experience I recommend using this mod alongside mods that remove the healing effect from food, water and sleep and make stimpaks much rarer. You should be able to find them inside the appropriate containers or buy them from chem vendors or doctors. bandages are more common and surgery kits are rare. First aid kits are the hardest to find. This is a work in progress and I'm not done yet, ultimately I want to be able to feel some of the pain and agony the lone survivor feels after being shot or attacked by a monster by adding sound effects and animations. (some engine limitations have slowed me down) Future plans Expanding the effects More animations. Compatibility Load last in your load order Compatible with VIS Not Compatible with Horizon (there is a patch to stabilize your game if you want to use both) Before Installing this mod back up your saves. Uninstall Make sure you have no active effects (e.g. cardiac arrest, blood loss and etc.) Vanilla effects or effects added by other mods are fine.

So you must be careful from them. If you have other infection in your body from long time then they can take this dangerous shape. This video provides photos and narrator interpretation of Blood Agar (BAP) specialized bacterial growth medium. Examples of sterile BAP as well as plates with growth of normal flora (gamma and alpha hemolytic bacteria) are contrasted with the presence of beta hemolytic Streptococcus pyogenes. This video provides complete information on how to determine if bacteria exhibiting beta hemolysis on BAP are Streptococcus pyogenes. There are other types of bacteria that are beta hemolytic, however S. pyogenes will grow in punctiform (tiny round) creamy white colonies. S. pyogenes is a Gram-positive, beta hemolytic bacteria. There are also Gram-negative beta hemolytic bacteria, but their colony morphology is different than that of S. pyogenes.
